Standard bacterial indicators like Enterococcus and E. coli confirm contamination is present but cannot identify whether the source is human or animal without additional faecal source tracking analysis, such as testing for the presence of human Bacteroides. Faecal source tracking (FST) is the process of determining whether faecal contamination detected in water originates from human or non-human sources. Standard faecal indicator bacteria testing confirms contamination exists but cannot distinguish between a human sewage leak and animal runoff. Source tracking uses host-specific genetic markers to make that distinction. The most established human-specific marker is the HF183 region of the 16S rRNA gene, found in Bacteroides dorei, a bacterium highly abundant in the human gut microbiome, but not in other animals. Identifying the contamination source allows councils and water managers to direct remediation efforts accurately, inspecting sewer infrastructure when human markers are present rather than pursuing costly and misdirected responses.
